PENGARUH PERBEDAAN KONSENTRASI PELARUT EKSTRAKSI TERHADAP AKTIVITAS ANTIOKSIDAN EKSTRAK DAUN Rhizophora mucronata
ABSTRAK
Tanaman mangrove, terutama Rhizophora mucronata, terbukti memiliki
profil FTIR (Fourier Transform Infrared Spectroscopy) ekstrak daun
Rhizophora mucronata.
antioksidannya semakin baik. Spektrum FTIR menunjukkan bahwa sampel
mengandung gugus fungsi O-H, C-H stretching sp?, C=C, CH? bending, CH?
Perbedaan konsentrasi pelarut berpengaruh
terhadap aktivitas antioksidan ekstrak daun Rhizophora mucronata. Ekstraksi
dengan pelarut etanol 96% direkomendasikan untuk menghasilkan aktivitas
antioksidan yang paling optimal.

ABSTRACT
Mangrove plants, particularly Rhizophora mucronata, are known to possess
strong antioxidant activity capable of neutralizing free radicals and reducing
the risk of diseases such as cancer, heart disease, and other degenerative
conditions. This antioxidant activity is influenced by the presence of bioactive
compounds, whose extraction depends on the concentration of the solvent used.
This study aims to determine the effect of different ethanol solvent
concentrations on the antioxidant activity and FTIR (Fourier Transform
Infrared Spectroscopy) profile of Rhizophora mucronata leaf extract. Samples
were obtained through sonication extraction using ethanol at concentrations of
50%, 70%, and 96%. Antioxidant activity was measured using the DPPH (2,2-
diphenyl-1-picrylhydrazyl) method and statistically analyzed with the
Kruskal?Wallis test using SPSS software. The FTIR profiles were analyzed
using PCA (Principal Component Analysis) through the PAST 4 application.
The results showed IC50 values of 33.97 ? 0.15 ppm, 22.49 ? 2.02 ppm, and
18.17 ? 0.52 ppm for the 50%, 70%, and 96% ethanol extracts, respectively,
with a significance level of p ? 0.05. These results indicate that lower solvent
polarity leads to better antioxidant capacity. The FTIR spectra revealed the
presence of functional groups including O?H, C?H stretching (sp?), C=C, CH?
bending, CH? bending, C?O alcohol, C?O ether, and C?Br. PCA analysis
showed that the 96% and 70% ethanol extracts had similar spectral
characteristics, while the 50% ethanol extract exhibited distinct differences.
Therefore, solvent concentration significantly affects the antioxidant activity
and chemical composition of Rhizophora mucronata leaf extracts. Extraction
using 96% ethanol is recommended to achieve optimal antioxidant activity.
