Cutibacterium acnes lipase (PDB ID: 6KHM). Analisis dilakukan secara in silico
interaksi ligan-reseptor. Evaluasi sifat fisikokimia dilakukan menggunakan
SwissADME, sedangkan prediksi profil ADMET dianalisis melalui pkCSM.
senyawa terbaik yaitu 2-(1H-indol-3-yl)-3-[4-(trifluoromethyl)phenyl]
acrylonitrile, isokuersetin, dan jubanin C. Analisis sifat fisikokimia menunjukkan
bahwa 2-(1H-indol-3-yl)-3-[4-(trifluoromethyl)phenyl]acrylonitrile memenuhi
kriteria kelayakan obat secara oral dengan parameter: berat molekul 316,32 Da;
refraktivitas molar 89,35; jumlah rotatable bond 4; log P 2,51; hydrogen bond

ABSTRACT
Acne vulgaris is a chronic inflammatory skin disease commonly experienced by
adolescents worldwide. The use of antibiotics as the main therapy often leads to
resistance, highlighting the need for alternative treatments based on natural
products with a lower risk of resistance. Caulerpa lentillifera is a green alga rich in
bioactive compounds such as flavonoids, phenolics, and fatty acids. These
compounds are known to possess antibacterial activity and potential anti-acne
properties. This study aimed to predict the potential of bioactive compounds from
Caulerpa lentillifera as anti-acne agents through inhibition of the Cutibacterium
acnes lipase enzyme (PDB ID: 6KHM). The analysis was conducted in silico using
YASARA Structure with a force-field?based scoring function and a flexible?rigid
docking approach to evaluate ligand-receptor interactions. Physicochemical
properties were assessed using SwissADME, while ADMET profiles were
predicted using pkCSM. The results showed that several bioactive compounds from
Caulerpa lentillifera exhibited good binding affinity toward Cutibacterium acnes
lipase, with three top candidates identified as 2-(1H-indol-3-yl)-3-[4-
(trifluoromethyl)phenyl]acrylonitrile, isoquercetin, and jubanine C.
Physicochemical predictions indicated that 2-(1H-indol-3-yl)-3-[4-
(trifluoromethyl)phenyl]acrylonitrile met the criteria for oral drug-likeness, with
the following parameters: molecular weight 316.32 Da; molar refractivity 89.35; 4
rotatable bonds; log P 2.51; 2 hydrogen bond donors; 4 hydrogen bond acceptors;
and a topological polar surface area of 35.88 Ų. Meanwhile, ADMET prediction
suggested that isoquercetin demonstrated a relatively better safety profile compared
to the other candidate compounds.
